I am not the primary operator of the 4700 but I do not believe we calibrated the plate in the 4700 software. Does the imaging software use this calibration? During setup I find the edge of the sample (usually the edge of the double sided tape) and record the absolute x/y values. I then use these numbers in the imaging software to set the area for scanning. The next time I run I will try to observe if the effect is systematic or not.

I would like to try to get resolution into the 20-40 um range. The raster on the 4700 goes down to 10um i believe. Do you typicaly fix standard “spots” of whatever analyte you are looking for? Recently I started adding spots next to the sections with a std prep of the neuropeptide of interest.
